[Abstract]Objective:ToobservetheeffectofelectroacupunctureontheexpressionofHtrA1inastrocyteswithinthe hippocampaltissueofratswithacutecerebralinfarction.Methods:Twenty-fourmaleSDratswereselectedandrandomly divided intothesham-operation,model,andelectroacupuncturegroups,with8ratsineachgroup.Themodel and electroacupuncturegroupsunderwentrightmidlecerebralarteryoclusion(MCAO)modelingviatheintraluminal filament occlusionmethod.Thesham-operationgroupreceivedidenticalsurgical exposurewithout filamentinsertion.Twenty-fourhours aftersuccesful modeling,ratsintheelectroacupuncturegroupreceivedelectroacupuncture treatmentatBaihui(GV2o)and Shenting(GV24)using dense-disperse waveswithalternating frequencies of2/15 Hz andacurrent intensityof 1.5–2 mA. Cerebralblodflowchangesastrocyteexpressionandactivationstatus,HrAlmRNAlevels,HtrAlproteinexpressonlevels, and TNF- α ,IL- 1β ,and IL-6 levels of three groups were measured and compared seven days after the operation.Results: WesternblotingshowedthatHtrA1expressioninthemodelgroupwassignificantlyincreasedcomparedtothesham-operation and electroacupuncture groups(both P<0.05 ).Quantitative real-time PCR (qRT-PCR)showed that HtrA1 mRNA expression level in the electroacupuncture group was significantly lower than that in the model group( P<0.05 ).ELISA showed that the model group had the elevated levels of TNF- α ,IL- 1β ,and IL-6 compared to the sham-operation group (all P<0.05 ),while the electroacupuncture group had the reduced levels of TNF- α,lL-1lβ ,and IL-6 compared to the model group(all P<0.05 ). Conclusions:lectroacupuncturecanleviateijuryproerologicalfuctioancerebralbloodflondpst inflammatoryresponseandthemechanismmaybeassociatedwiththeinibitionof HtrA1proteinexpressionandsuppressonof astrocyte polarization.
