连翘苷脂质体凝胶的制备工艺优化及其质量评价

Abstract:Using forsythin as the main raw material,forsythin liposomes were prepared by the film dispersion ultrasonic method,and thephysical and chemical propertiesof the prepared liposomes were explored.Furthermore,the forsythin liposomeswere developed into forsythin liposome gel.Taking the appearance,viscosity,spreadabilityand centrifugabilityof the gelas the evaluation indexes,based on the single-factor test,the preparation process of the forsythin liposome gel was optimized with the response surface method.The appearance of the forsythin liposome gel was evaluated by the visual inspection method,and the pH value of the forsythin liposome gel was determined by a pH meter.The centrifugal stability,heat resistance stability and cold resistance stabilityof the forsythin liposome gel were systematically investigated.The results showed that the forsythin liposomes presented arelatively regular spherical profile,with a particle size of( 123.4±1.6) nm,a PDI of 0.220±0.019 ,and a Zeta potential of (-24.63±0.71)mV .The optimal process conditions for the forsythin liposome gel were determined as follows:the dosage of Carbopol-940 was 0.75% ,the dosage of triethanolamine was 1% ,and the dosage of glycerol was 10% . The prepared forsythin liposome gel was a smooth,shiny,uniform and stable semi-solid with moderate viscosity and extremely easy spreadability. The average pH value of the forsythin liposome gel was 6.9±0.05 ,which met the requirements for topical skin preparations.Meanwhile,the gel had good centrifugal stability,heat resistance stability and cold resistance stability.
