白灵菇蛋白质的分级提取及其性质研究

Abstract:Pleurotus nebrodensis was used as raw material to extract albumin and protein isolate by acid precipitation method. Then,globulin,prolamin and glutelin in Pleurotus nebrodensis protein isolate were extracted by Osborne method,and their physical and chemical properties and functions were studied.The results showed that albumin,globulin,gliadin and glutelin accounted for 62.24% , 3.57% , 2.42% , 21.77% (2 of the total protein inPleurotusnebrodensis,respectively.The isoelectricpoints of the four proteins were 4.0,4.5,5.0,5.5,respectively.The contents of albumin in the albumin lyophilized powder was 71.63% , and the contents of gltolin in the glutelin lyophilized powder was 74.49% after purification by adjusting the isoelectric point.Albumin and glutelin had the best solubility at pH9,which were 71.2% and 76.7% The water holding capacity of albumin and glutelin were 2.07g/g and 3.11g/g ,and theoil holding capacity were 4.87g/g and 3.97g/g ,respectively.In the range of pH7\~1O,both albumin and glutelinhad good foaming property,and thefoaming property increase with the increase of alkalinity.The foaming property of glutelin were better than those of albumin.The two proteins had good foaming stability between pH8~10 Key words: Pleurotus nebrodensis; protein; graded extraction; Osborne method; albumin; glutelin
