Effects of Cardiac Homing Peptide-Modified Liposomes Loaded with Colchicine on Cardiac Function Following Myocardial Infarction

【Abstract】ObjectiveTo construct liposomes loaded withcolchicineandmodified withcardiac homing peptide(CCP)and investigatetheirfectsoncardiacfunctionafermyocardialinfarction(M).MethdsCCPwerepreparedusingthethin-filmhydration method.ThemorphologyofCCPwascharacterizedbytransmissonelectronmicroscopyH9c2ratcardiomyocytehypoxiamodelswere establishedtoevaluatethco-loalzationofCPwithardioyoyes.Tecellviablitywasasessdusingthellountingit-8ssay. Cellaruptakeandco-lcalizionwereobseedbyconfocallasersaningmicopy.SDatseredvidedinto6groupsha,P, colchicine-loadedliposoes(C)colicine(COL),lposomes(L)andMTreatmentswereadministeredviatailveinictiofor4 weks.Echocardiogaphyasperfoedatwek4folowedbyhistopathologicaltaningofyocardialtisses.ResultsBothCCandCP exhibiteduniformsphericalmorphology.Theparticlesizeandpolydispersityindex(PD)ofCPwere(183.5±2.1)nmandO.1960.014, respectively,while CCP measured (232.2±8.0)nm and 0.299±O.O51.In vitro,CCP demonstrated higher cellular uptake than COL and CP, withconfocaliaingcofiingectiecardoyocygetingbardacoingpeptide.InioteCC,C,ndCOousod reducedolpstddeltdedolfiadoo revealedsignifcantlyateuatedmyocadialfrosisinthCCPgoup.Echocadiogaphyidicatedmarkediprovementinadacfuction parametersintheCCP,CP,andCOLgroupsversustheMgoup.ConclusionCCPeectivelytargethypoxiccardiomyocyes,viate myocardial fibrosis,and improve cardiac function post-MI,providing anovel therapeutic strategy for MI treatment.
