[ABSTRACT]ObjectiveTo investigate the influenceof extracelular matrix (ECM)stifnessontheexpressionof inflammatoryfactorsinprimary humanprostatecancercels.MethodsPrimary human prostatecancercellswereinoculated intoculture dishes pre-incubated with 0.1g/L ECM solution (group A) or 8g/L ECM solution (group B),and bright-field microscopy was usedto observe cellmorphology.Onday4ofculture,RT-qPCR and Western blot were used to measure therelative mRNA and proteinexpressionlevelsoftheinflammatoryfactorsC-Cmotifchemokineligand2(CCL),C-X-Cmotifchemokineligand10 (CXCL1O),intercelularadhesion molecule-1(ICAM1),andinterleukin-32(IL-32)incell.ResultsBright-fieldmicrosopy showedthatat24oursafterinoculation,thecellsingroupAgraduallyaggregatedandformedsht-likestructures,whilethose in groupBexhibited multilayeredcellaggregates.RT-qPCR showed thatcompared with groupA,group Bhadsignificantlyhigher relative mRNA expression levels of the inflammatory factors CCL2,CXCL10,ICAM1,and IL-32 ( t=5.30-16.23,P<0.0 5). Western blot showed thatcompared with groupA,group Bhad significantly higher relative protein expresion levelsof ICAM1, IL-32,and CCL2 (t=2.78-14.72,P<0.05) ,while there was no significant difference in the relative protein expression level of CXCL10 between groups A and B( P>0.05 .Conclusion Soft ECM substrate can promote the expression of the inflammatory factors CCL2,CXCL1o,ICAM1,and IL-32 in primary human prostate cancer cells.
