【AbstractObjective:Toinvestigate teinterventionefectandactivemehansmof taditionalChinesemedicine Masdeniatencis

simaoncolitis-associatedcolorectalcancer(CAC).Methods:Hig-resolutionmasspectrometryanda precisemassnumberdata

basewereusedtoidentifythechemicalcomponentsofMarsdeniatenacissmandnetworkparmacologymethodsereusedtobtain

themainactivecomponentsandtargetsofMarsdeniatenacisimainthetreatmentofcolorecalcancer(CRC)andanalyzeitsmeh

nismofactionagaistCRC.AousemodelofCACwasestablisd,ndtenMarsdenitenacisimaasanaleditesfitsefct

ondisease activity index(DAI),body mass,colorectal morphology,and serum levels of tumor necrosis factor- α (TNF- α )and interleu

kin-1β(IL-1β)inCAC mice.Furthermore,6SrDNAsequencing techniquewasused toassesstheimpactof Marsdeniatenacissima onthediversity ofintestinalmicrobiotainmice.Results:Atotalof 50 componentswere identified bymass spectrometry,and 37 active componentswere identified bynetwork pharmacology.Furthermore, 313targetsrelated to CRCwereidentified.A totalof 958GO terms and 173 KEGG terms were obtained,among which inflammation, oxidative stress,and cancerpathways showed arelatively high degreeofassociationwith Marsdenia tenacissima.After modeling,the micein each group had the manifestationssuchasdiarrhea,bloody stool,prolapsefsdweightlssndteodelgoupadaigicantcreaseinorfeachcleilete MarsdeniatenacissimgrouphdsignicanteductioinDsoreofthemodeliceafterthefirstecondndtdles( P= 0.007, P<0.001 ,and P<0.001 ). In addition,compared with the model group,the Marsdenia tenacisima group had a significant increaseicolortalnghllviatiofitestalselling,elativelactructureoielilels,ducoiato cellinfilratoteainbeluiitalplsdntuisiesf TNF- α and IL-6 in CAC mice( P<0.001 and O.Oo2).The Marsdenia tenacissima group had significant reductions in the abundance anddiversityofintestialmicrobotaandcomparedwithteodelgoup,theMarsdeniatenacisimagouphadsignificantduction intherelativeabundanceofBacteroidesandsignificantincreases intherelativeabundancesofMuribaculaceaeandClostridia-UCG014 ( P=0.002 andO.009).Conclusion:Marsdenia tenacisima exertsaninterventioneffectoncolorectalcancerin CAC mice,possibly byregulatingthepathways associated with inflammationandcancerand altering thediversityof intestinal microbiota.
