【AbstractObective:TinvstigatehtherintratuoralGemellahemolysans(Gamolysans)enanesteficacyoadjuvant
chemoimmunotherapy(NACI) inesophageal squamous cellcarcinoma(ESCC)-bearing micetheunderlying mechanism.Methods:
Twenty-five 4-week-old female C57BL/6 mice were romized into five groups( n=5 per group) : negative control(NC) group,chemo
therapy group,G.haemolysans + chemotherapy group,NACI group, G.haemolysans + NACI group. A germ-free mouse model was
establishedineachgroupbyadministeringantibioticcocktailfor2weeks.OndayO,ECC-bearing mousemodelswerebuiltthrough
subcutanous inoculation ithermurine esophagealcancercelline AKRor G.haemolysans-AKRcels.Onday7whentumors
formedundertheskin,drugswereinjectedintotheabdomenthricewekly.Onday28whenthetreatmentswerefinished,themice
weresacrificed to harvest their tumors, then the tumors were fixed in 4% paraformaldehyde.We locatedquantifiedG.haemolysans
withinthetumorsthoughfuoresceceinsituhbridizatio,ndcharacterzedimunecellompositioninthetumorimmuneo
vironment(TIME)themicetreatedwithNACIthroughmultipleximmunohistochemistryflowcytometry.Results:G.haemolysans
significantly strengthened the efficacy NACI in ESCC( P<0.05 ),whereas no chemosensitizing effects were observed on chemo
therapy in ESCC-bearing mice.G. haemolysans significantly enhanced natural killer(NK)cell infiltrationwithin the TIME( P <0.01).
Conclusion:G. haemolysans improves NACI efficacy in ESCCbearing micebyincreasingNKcell recruitmentin the TIME.
