Abstract:ObjectiveTo investigate the efectsevlurane onneuroinflammation inrats with spinal cord injury by regulating thereactiveoxygenspecies(ROS)/thioredoxin-interactingprotein(TXNIP)/nucleotide-bindingoligomerization domain-like reeptor protein 3(NLRP3)pathway.MethodsSeventy-two rats were randomly divided into six groups using arandomnumber tablemethod,with12rats inach group:the modelgroup(establishing theratmodelspinalcord injury), the sham operation group,the low-,medium- and high-concentration sevlurane groups ( 1% , 2% and 4% sevlurane, respectively) and the combination group ( 4% sevlurane + 3.7 mg/kg reactive oxygen species (ROS) activator trimethylamine N-oxide).Spinalcord injurybehavior (BBB)scoreand footprint imprint testwereused to evaluate motor function.ELISA method was used to detect the levels IL-18,TNF- α and IL- ⋅1β in spinal cord tissue.HE staining was used to detect pathologicalchanges in spinal cord tissue.TUNELmethod wasused todetect cellapoptosis.Inaddition,the expresion TXNIP and NLRP3 mRNA were detected byqRT-PCR.ROSactivity was detected by ROS kit.Western blot assay was used to detect the expression ROS/TXNIP/NLRP3 pathway proteins.ResultsThe sham operation group showed normal gait, normal cell morphology with orderly arrangementand intact spinal cord structure.The model grouppresented with toe dragging,severe spinal cord tissue injuryand inflammatorycellinfiltration.TheBBB score was lower inthe model group compared to that the sham surgery group,while IL-18,TNF- ∇⋅α∝ , IL-1β levels,apoptosis rate,ROS activity,TXNIP, NLP3 mRNA and protein expression were increased ( P<0.05 ).The above indexes were improved in the low-concentration, medium-concentration and high-concentrationsevlurane groups compared with thosein the model group.However,the combined groupreversedtheimprovement results the high-concentration sevlurane group.ConclusionSevlurane can alleviate neuroinflammation in rats with spinal cord injury by inhibiting the ROS/TXNIP/NLRP3 pathway.
