Abstract: Inodertoclarifythe biochemicalfunctions oftobaccoendophyticbacteria and explore theirapplication intobaccoleaf fermentation,ain4ithgfciecyigraingproeiscellos,achlgn,ndpctiassoatedfrofr tobaccoplants.Basedonmorphologicalcharacteristicsandphylogenetictreanalysis,NS42waspreliminarilyidenifidasBailus velezensis.FolowingfermentationwithNS42,thechemicalcompositionoftobaccoleaveswasmodulated,leadingtoan enhancementinthromaqualityflue-uredobacolaves.Notablybothtearomaitesityadsweeessoffue-uretobac leavesincreasedhileitaionndnsiveorsereimsedsultinginmorepleaantftertastehducinggar content increased by 18.51% The contents of starch, cellulose, lignin, protein, and pectin in tobacco leaves decreased by 15.61% 9.67% , 8.49% ,12.70,and 16.47% ,respectively. The content ofaroma compoundsincluding5-methylfurfural, β. dihydrodamascenone,megastigmatrienone,farnesylacetone, β -ionone,solanone,phenylethyl alcohol,benzaldehyde,and neophytadiene were significantly elevated.Thewhole genomeofNS42comprises 6885670540base pairs encoding 3919 genes, among which7enswererelatedtooteinegadation,6toellosedegradation6ttarchegadation9topectiegadtio and4tolignindegradation.Thisstraincanutilizevarious plysaccaridesandproteinsascarbonandntrogensources.Inthisstudy anendophyticBacilusstrainwithobustdegradingenzymeactivitywasisolatedandcharacterized,demonstratingasignificant impactonthechemicalaromacomponentsandsensoryqualityof tobaccoleaves,therebyshowcasingsubstantialpotential for enhancing tobacco leaf quality.
