Screening and Identification of Endophytic Bacillus Velezensis in Tobacco and Its Application in Tobacco Leaf Fermentation

Abstract： Inodertoclarifythe biochemicalfunctions oftobaccoendophyticbacteria and explore theirapplication intobaccoleaf fermentation，ain4ithgfciecyigraingproeiscellos，achlgn，ndpctiassoatedfrofr tobaccoplants.Basedonmorphologicalcharacteristicsandphylogenetictreanalysis，NS42waspreliminarilyidenifidasBailus velezensis.FolowingfermentationwithNS42，thechemicalcompositionoftobaccoleaveswasmodulated，leadingtoan enhancementinthromaqualityflue-uredobacolaves.Notablybothtearomaitesityadsweeessoffue-uretobac leavesincreasedhileitaionndnsiveorsereimsedsultinginmorepleaantftertastehducinggar content increased by 18.51% The contents of starch， cellulose， lignin， protein， and pectin in tobacco leaves decreased by 15.61% 9.67% ， 8.49% ，12.70，and 16.47% ，respectively. The content ofaroma compoundsincluding5-methylfurfural， β. dihydrodamascenone，megastigmatrienone，farnesylacetone， β -ionone，solanone，phenylethyl alcohol，benzaldehyde，and neophytadiene were significantly elevated.Thewhole genomeofNS42comprises 6885670540base pairs encoding 3919 genes， among which7enswererelatedtooteinegadation，6toellosedegradation6ttarchegadation9topectiegadtio and4tolignindegradation.Thisstraincanutilizevarious plysaccaridesandproteinsascarbonandntrogensources.Inthisstudy anendophyticBacilusstrainwithobustdegradingenzymeactivitywasisolatedandcharacterized，demonstratingasignificant impactonthechemicalaromacomponentsandsensoryqualityof tobaccoleaves，therebyshowcasingsubstantialpotential for enhancing tobacco leaf quality.
