Fermentation Optimization and Application of High-Yield α-amylase Genetically Engineered Bacterium Strain A8-1

Abstract: In order to improve the a -amylase production activity of genetically engineered strain BAX-5/PT-17 SPo02amy(A8-1), improveitsindustrialapplcabilityandapplicationeffectintobacoprocessng,aseresof parameters inthefermentationproce wereoptimizedthroughsinglefactortestandresponsesurface experiments,andthescaled-upprocesswasvalidated inafermenter. Meanwhile,changes in chemical composition and sensory quality of tobacco with a -amylase treatment were analyzed. After optimization,the optimal fermentation medium and conditions for strain fermentation were determined as: corn meal 65g/L, soybean cakemeal 70g/L ,NaC1 10g/L , CaCl2 (204 3g/L ,fermentation time 52h ,temperature 40°C , pH6.0 ,inoculation volume 3% .The optimized a -amylaseactivityreached 1880.1U/mL . Compared with 30.6U/mL before optimization,the enzyme activity increased by 61.4-fold changes, and the enzyme activity could achieve 2276.2U/mL in 5L fermenter. After 7U/g (2号 a -amylase treatment, the starch content of tobacco decreased by 19.9% ,the total sugar and reducing sugar content increased by 8.09% and 7.81% ,respectively. The aromaprecursorsubstancesandotherindicatorswerepositivelyimprovedinvaryingdegres.Thetreatedtobaccoexhibitedenanced aromarichness,mootherandmildersmoke,withreducediitancyndof-otes.Theoptimedfermentationprocessignificantly improvedtheenzyeyieldofgeneticallyengineredstrainA8-1,demonstratingstrongindustrialaplicationpotential.Its implementation in cigarete production effectively modifies chemical composition and elevates smoke quality.
