Identification and Evaluation of Antiviral Components in Glandular Trichome Secretions of Sun/Air-cured Tobacco

Abstract: Inordertoeucidate thecontrolefectofglandulartrichomesecretionsfromsun/air-curedtobacoonplantviradiseases andtoidentifysuperioractivecompoents,thissudyassedtheanti-MVactivityfethanolextractsfromglandulartichomesf 28 sun/air-cured tobaccogermplasmresources using asaprubing-inoculation method.Inadition,thecomposition andcontentof diterpenoidsandsucroseesters (SEs)intheglandulartrichomesecretionsweredeterminedbyultra-high performanceliquid chromatography(UPLC)andgaschromatographymassspectrometry(GCMS).Corelation,partialleastsquares,andcustering analyseswereusedtoinvestigatetherelationship between thecontentof glandular trichomesecretionsandtheanti-TMVactivity. Theresultsshowedthat: (1)Theethanolicextractsof glandular trichomesfrom19germplasmsamples exhibited antiviral activity against TMV, with Q16 (Meitan sun-cured tobacco) showing the best virucidal efect, inhibiting necrotic spots by 68.87% ; (2) There wasa significant or extremely significant negative correlation between cis-abienol and a -cembratriene-diol, β -cembratriene-diol,EI, andSEII.Coveselyisldsiantorelysiatpoieoelaoit,SVd importantlycbol,Ediedgligianposiieelaioiteoateoict. (3)Cis-abienol,SE,andEVrentifiedasteajoactiveomponentsagaistinteladulartrcometisof sun/air-cured tobacco.Additionally, the validation experiments showed that cis-abienol (200μg/mL) and SEs (300μg/mL )exhibited themost antiviral activities,with necrosis inhibition rate of 44.32% and 53.19% respectively.These rateswere significantly higer than those achieved by a plant-derived pesticide ( 0.5% amino-oligosaccharides).The research results provide a material foundation and technical supportforthe efficient utilization and the development of plant-derivedantiviral activecomponents.
