Abstract:Toidentifyhigh-qualitycigartobaccogermplasmresistanttobacterialwilt(Ralstoniasolanacearum)andprovide molecular markersforsubsequent breeding,this studyutilized GBS (genotyping-bysequencing)resequencingdatafrom19tobaco germplasm resources,including the cigar tobacoline ZX145.Thenumber of identified ZX145-specific SNP loci was 221.The specific SNPloci were screened forthe development of KASP(kompetitivealele-specific CR)molecularmarkers.Validationusing 71 tobacco germplasm resourcesof diversetypes yielded 28KASP marker pairscapable of distinguishing ZX145 from other germplasm.AspecificSNPfingerprintforZX145wasconstructedusing24lociselected tocoverallchromosomes.Fieldevaluations forbacterialwiltsistane,alogsidecomprehesiveasesentsofgronomicandyieldtraitsereonductedindiseasenser ofFujian,Guizhou,andAnhui Province.TheresultsshowedthatZX145exhibited moderatelyresistance-resistanceinthedisease nurseries,whichwassuperiortotheconventionalcontrolvarieties,anditsoutputvalueperformedbeter.ConsequentlyZX145 represents an eliteresistant germplasmresource forbreding bacterial wilt-resistant cigar tobaccocultivars.The developed KASP markers provide robust molecular tools to support its utilization in breeding programs.
