Abstract:Toexplore themolecularcharacteristicsofBMP4inPelodiscus sinensisanditsroleinfoliculardevelopment,we employed molecularcloningand bioiformatics methodstoobtaintheful-lengthcDNAoftheBMP4 gene in this species.Furthemore, quantitativePCR wasemployedtoaalyzethetissue-specifcexpressonofthisgeneandtheexpressonpates intheovaryduring thereproductivecycleandtheembryonic gonads.Theful-engthcDNAsequenceofthis genewas1743bp,encompassinga45-bp 5'-UTR,a75-bp3-UTR,anda1209-bpopenreading frame (ORF)that encoded40aminoacidresidues.Thededuced protein had the isoelectric point (pI) of 9.24 and the molecular mass of 45.07kD .The amino acid sequence of BMP4 contained a signal peptide composedof24aminoacidresiduesand6helicalstructures.Thephylogeneticanalysisoftheaminoacidsequencesshowedthat te BMP4 of P. sinensisclusteredwiththoseofChelonia mydas,Carettcarett,andDermochelyscoriacea.ThemRNAlevelofBMP4 in the ovarian tissue washigher than those in the liver,muscle,intestine,and testis tissuesamples (P<0.05) .During thereproductivecycle of P. sinensis,thelowestmRNAlevelofBMP4wasobservedinwinter,whilethehighestmRNAleveloccurredduringthefollcular developmentstage inspring.Followingovulation,themRNAlevelofBMP4declined.Theresultssuggst hatBMP4isinvolvedin regulating the growth,maturation,and ovulation of ovarian follicles in P. sinensis.Inaddition,BMP4 may be involved in embryonic gonad development, but not in embryonic gonad differentiation.
