Abstract:Thelateembryogenesisabundant protein (LEA)familyplaysasignifcantrole inseeddevelopmentandstressresistaceof plants.This studyemploedbioiformaticstoolsandthesubcellularlocalizationmethod toexplorethetructureandfunctionofLEA familymember,HvnLEA6,inHordeumvulgareL.var.nudumHook.f.TheresultsidicatedthatthepromoterregioofHnLEA6 contained multiple elements related toabscisicacid (ABA),jasmonicacid (JA),uxin,andsed-specificresponses.HA6 consistedofl17aminoacidresidues,washydrophilic,lackedasignalpeptideandtransmembranestructureandwaslocaledinthe nucleus.In the secondary structure ofHvnLEA6, a -helicesandrandom coilsaccounted for 62.39% and 37.61% ,respectively,with no extended strands or β -sheets.HvnLEA6possessedaconservedLEA_1domain,whichwasan a -helixandrepresentedthemost conservedstructureinHvnLEA6.Theproteinsurfaceregions withpositiveandnegativechargeswere alsoprimarilylocatedwithin theLEA_1domain.Therefore,thisdomain wasconsidered themain functionalregionofHvnLEA6.Moleculardynamicssimulations have revealed that HvnLEA6 exhibited high structural flexibility in the transition zone (aa75-85)between a -helices and random coils. Inconclusion,HvnLEA6maybedirectlyinvoledinprotectingthenucleicacid-proteinstructures withinthenucleusagaiststress conditions and the activity or expression is jointly regulated by ABA and JA.
