Abstract:Tostudythefunctionofthevoltage-dependentanionchannel gene (VDACinSpodopterafrugiperda,thisstudyclonedS. frugiperda VDAC(SfVDAC)andusedbionformaticstolstoanalyzethisgene.Furthermore,R-qPCRwasemployedtoquantifythe relative expression levels of SfVDAC in the eggs,larvae,pupae,and adultsof S.frugiperda.The results showed that the open reading frame(ORF)sequenceofSfVDACwas849bpinlength,encoding282aminoacidresiduesandcontainingonePorin_3domain.The deduced protein had a molecular weight of 30.0kDa and an isoelectric point of 7.74. No signal peptide or transmembrane structure wasidentifed,andtheproteinwaspredictedas hydrophilic.Multiplesequencealignmentandphylogeneticanalysisidicatedthat SfVDAC wascloselyrelatedtotheVDACsofother lepidopteraninsectssuchas Spodoptera lituraandS.exigua,and thehomology Wasrelativelyhigh (97.02%) .The secondary structure prediction of SfVDAC showed that the most abundant structure was random coil (35.82%) ,andthe leastwas β -turn (6.74%) . The tertiary structure analysis revealed that the protein had two channel structures. The results also showed that, in terms of developmental stages,the expression level of SfVDAC Was the highest in the larvae,while it Was the lowest in the eggs. Additionally, the tissue expression results indicated that SfVDAC was highly expressed in the midgut and Malpighian tubules,while the expression level was the lowest in the hemolymph.
