Abstract: Phytoene desaturase (PDS),which affects the biosynthesis of carotenoids,is a key enzyme in the synthetic pathway. In order to study the function of the PDS gene of Leymus chinensis,in this experiment,the PDS gene was successully cloned from‘Zhongke No.1’L. chinensis and bioinformatics analysis was performed.Protein structure analysis,subcellular localization,transcriptional self-activation detectionand expression pattern analysis were conducted as well. The results showed that the ful-ength cDNA sequence of this gene is 1743bp ,encoding 58O amino acids. Chloroplast transit peptides may exist in the LcPDS protein,and thethe protein has the closest relationship with thePDS protein of wheat(Triticum aestirum).Subcellular localization revealed that the protein is localized in thechloroplast.The transcriptional self-activation test showed that LcPDS does not have self-activation. There was no significant difference in the LcPDS gene expression among different developmental stages of leaves.The expression levels in various organs followed the order:root > leaf > stem( P<0.05) .The LcPDS gene is regulated by gibberellin,abscisic acid,methyl jasmonate and salicylic acid. This study provides a scientific foundation for further research on the function of the LcPDS gene.
