Roleand mechanistic study of the miR-194-5p/UBA2 in the progression of clear cell renal cell carcinoma

Abstract：Objective：To investigate the expression patern of miR-194-5p inclear cellrenal cellcarcinoma （cRCC） anditseffectsonmalignantbiologicalbehaviorsof tumorcels，andtovalidatethetargetedregulatoryrelationshipbetween miR-194-5p and UBA2.Methods：The expression of miR-194-5p inccRCC tssues and celines was measured by real timeRT-PCR.miR-194-5pagomir，miR-194-5pantagomirandcorrsponding negativecontrolwere transfected into786-O and Caki-1cells，and theexpression of UBA2 protein and mRNA was detected.Cell proliferation，migration/invasion， andapoptosis were evaluated using CCK-8assays，transwellassys，andflow cytometry，respectively.Adual-luciferase reporter assay was performed to verify the direct binding between miR-194-5p and the 3′ UTR of UBA2. Results ： miR-194- ⋅5p was significantly downregulated in ccRCC tisses and ccRCC cellines. Up-regulation of miR-194-5p could reduce theexpresionof UBA2 proteinand mRNA，inhibitcellproliferation，migrationand invasion，andpromote apoptosis.Thedual-luciferasereporterassayshowed thatmiR-194-5pspecificallysuppressed thereporteractivityof constructs containing the wild-type UBA2 3'UTR，whereas no significant suppression was observed in UBA2 3'UTR （2 5′ -UGUUACA-3'mutant constructs.Conclusion：Restoration of miR-194-5p expresion can suppress the malignant biological behaviors of ccRCC cells by directly targeting and inhibiting UBA2.
