Abstract: ObjectiveTo improve the method establishing glioblastoma organoids(GBO) and explore the feasibility evaluating the effect new treatments.MethodsThe postoperative pathological tissues glioblastoma(GBM) diagnosed as WHO grade IV were collected,and the tumor organoids and primary cells from different patientswere established and compared with the parental tumors,and the GBO were used to evaluate the efect tumor-treating field treatment in vitro.ResultsFifteen the eighteen GBM tissues were successfully established and retained the structural characteristics the parental tumors,which could be cultured for a long time and used for experiments. Primary GBM cells were obtained during the process,and expressed CD133 in immunluorescence staining.In vitro experiments confirmed that TTF treatment could inhibit the growth GBO. ConclusionsThis study improved a low-cost method to rapidly establish GBO to evaluate the effect tumor-treating field treatment.
