Abstract:ObjectiveTooptimizetheextractionprocessoftheactivecomponentsofRadixPaeoniaeAlbaandassessthe antioxidantanti-inflammatoryandpigmentatio-reducingefectsofRadixPaeoniaeAbaextract(BT).Methodsoleular dockingidentifiedgalicacid,liquiritinandpeoniflorinaspotentialtyrosinaseibitorsandtheirontentsinwere determinedwithhigh-performanceliquidchromatography(HPLC).Basedonthecontentsofthe3inibitors,theirtyrosinaseinhibitingactivityandDPPHradicalscavengingcapacitycalculatedusingentropyweightmethod,theextractioncoditions Wereoptimizedforsolventratio,liquid-to-solidratio,extractiontime,andparticlesize.TheinhibitoryeffectsofTon tyrosinase activity and melaninwere assessed in α -MSH-treated B16F10 cells,and itseffect onreactive oxygen species (ROS) production was evaluated in H2O2 -treated HaCaTcells. Ina mouse model of melanogenesisinducedby UVB radiation, the effect of BST gavage (100, 150,and 200mg/kg )were evaluated by assessing skin conditions of the mice, Fontana-Masson staining,anddetectingserumlevelsofcytokinesrelatedtomelanogenesis,melanosometransport,metabolism,and inflammation. Results The optimized parameters for BST extraction included 50% ethanol, liquid-to-solid ratio of 15: 1, extraction time of 1h, and20-mesh particle size.BSTexhibited strong freeradical-scavengingactivity and significantly reducedmelanogenesisandincreasedtyrosinaseproteininB16cels,andloweredROSproductioninHaCaTcels.Inthe mousemodelsofmelanogenesis,BSTsignificantlysuppressdthemelanogenicenzymesandinflammatoryresponsesby reducing the levels of tyrosinase and TNF- α, thereby efectively reversed UVB-induced photoaging and hyperpigmentation. ConclusionTheoptimizedBSTextractionprocessisstableandpredictable.STshowsgoodskin-whiteningefect,whichis mediated posiblyby inhibitingtyrosinase activity andmodulating oxidativestressand inflammatorypathways.
