Abstract:ObjectiveToinvestigatetheefectoftotalAstragalussaponins (AST)onastrocyteswelinginducedbyglucoseoxygen deprivation and reoxygenation (OGD/R)and its molecular mechanism.Methods Primary astrocytes isolated from he brainof postnatalday2SDratswere treatedwithsolventvehicle,AST,orASTcombinedwithU-46619(ap38activator) 24h priortoandringexposuretoGD/R.Theangesincellumeandmorphologywereobsered,andintracelllaractive oxygen species (ROS)levelsand lactate dehydrogenase (LDH)activity were detected.The expressonof aquaporin 4 (AQP4) oncell membranewasobservedusing immunofluorescenceconfocalmicroscopy,and Western bloting wasperformedto detectthechanges inp38signaling pathwayphosphorylation levelsandAQP4 expresson level.ResultsASTtreatment significantlyreducedOGD/R-inducedincreaseinastrocytevolumeROlevelsandLDHrelease.TheAT-treatedcellsalso exhibitedimprovedcellmorphologyrducedaggregationofAQP4onthecellmembrane,anddecreasedp38phosphorlation andAQP4expression levels.TheprotectiveeffectsofASTagainst OGD/R-inducedcellinjuries were significantlyatteuated by combined treatment of the cels with U-46619. Conclusion AST alleviate astrocyte sweling induced by OGD/R injury by inhibiting the activation of the p38 signaling pathway and AQP4 overexpression on the cell membrane.
