Abstract:ObjectiveToinvestigatetheroleoftheAKT1/GLUT1signalingpathwayinmediatingtheinhibitoryefectofYiqi JieduFormula(YQJDF)againstnasopharyngealcarcinoma (NPC)cels.MethodsMoleculardockingwasemployedtoanalyze thebindingaityetweethactiecomponentsofadAK1.ssayaltecellalysis woud healingassayandMatrigel invasion chamber assay were used to evaluate theefect of YQJDF extract on proliferation, migration and invasionabiliesofumanNPCcellines5-8Fand6-1B,andthechanges incelular protein expressionlevels weredetected using Western blotting.Ina BALB/cnude mouse modelbearing NPCcellxenografts,tumor growth were observed following treatment with daily gavage with normal salineor YQJDF extract (15.357g/kg) for18 consecutive days or withintraperitonealinjectionsof5-Fueveryotherday.ThechangesintheexpresionsofAKT/GLUTsignalingaisproteins inthexenograftswereexaminedusingWestern bloting.Results IntheNPCcellines,treatment with YQJDFextract significantlidcllfgosdedpsof N-cadherinandvimentin.TeapplicationofGLUorAKactivatorspartialleversedtheibitoryefetsofQJDFon the NPC cels. In the tumor-bearing mouse models, treatment with YQJDF extract obviously suppresed tumor growth and down-regulated the expressionofAKT,p-AKTandGLUT1 inthetumor tissues.Conclusion YQJDF inhibitsproliferation, invasion,andmigrationof NPCcellsbyinhibiting the AKT1/GLUT1 signaling pathway.
