Abstract: Objective Toexplore the mechanism of Huayu Tongbian Decoction (HYTBD)forrelieving slowtransitconstipation (STC) inrats.Methods Sixty SDratmodels of STC induced by intragastric administration of loperamide hydrochloride suspension wererandomizedequallyinto5groupsfor treatment withdaily gavage withsaline (STCmodel group),low-, medium-,orighdoseHBD,ormosapride(positivecontrolgroup),withanother12normalratsreceivingsalinegavageas theblankcontrolgroup.Thfirstblackstooltime,fecalwatercontentandintestinalpropulsionrateoftheratsweremeasured, andthe ultrastructureofinterstitialCajal cels (ICCs)wasobserved using transmission electron microscopytocalculate the mitochondrialvacuolarrate.Thephagocytosisrateofmacrophages was detected usingimmunofluorescence assayandcolonic pathologieswereexainedsingHEstaing.eoloicexpresionlevelsof-KitCD47,3KandAktweredeectedith immunohistochemistryorWestenBlotingandtheircorrelationswitheferocytosiswereanalyzed.ResultsTheratodelsof STCshowed significantultrastructural damageoftheICCs withincreasedfirstblack stooltime,mitochondrialvacuolarrate, andcolonic expresionlevelsofCD47,p-PI3K/PI3Kandp-Akt/Aktproteis,loweredfecalwatercontent,intestialproulsion rateandc-Kitxpesonlel,itoutigiantangesingyicate.eatmentwithspeialleigh dose,significtlyrsdealteetstialsiategotosteolicKitpod decreasedthefistblackstooltime,mitochondrialvacuolarrateandcolonicexpreionsofCD47,p-PIK/PI3Kandp-AtAkt inthe mouse models.Conclusion HYTBDimproves STC inrats possibly by down-regulating colonic CD47 expressionand improving macrophagephagocytosisrateviainhibitingPI3KandAktphosphorylation,therebypromoting efferocytosisofthe ICCs.
