基于液相色谱-质谱串联法技术的32种类固醇激素的检测方法建立及其在妊娠全周期变化应用

Abstract:ObjectiveToestablishahighlysensitiveandspecificliquidchromatography-tandemmassspectrometry(C-MS/ MS)-basedanalyticalmethodfordetectingsteroidhormonesinhumanserum.MethodsSolid-phaseextraction (SPE)wasused forsamplepretreatment.Chromatographicconditionswereoptimized toachievebaselineseparation,andmassspectrometry parameters wereadjusted toensureaccurate detection of traceamounts of hormones.After validation of thelinear range, accuracy,and precision,the method wasapplied indetectionofserum samples from normalpregnant women foranalyzing dynamicvarationcaracteristicsf32seroidomosduringteist,cond,andtirdrimestersofpregnancyults The linear range of the method was 0.001-7500ng/mL, with a limit of detection (LOD) of 0.0007ng/mL . Both intra-day and inter-day coefficients of variation (CV) of the method were less than 10% ,the spiked recovery ranged from 86.21% to 112.67% the extraction efficiency was 85.05% 106.3%, and the matrix effects were 85.60%-113.22% . The results of detection using this methodfor serum steroid hormones duringpregnancyrevealed significant elevationof mostof thehormones(suchas11- deoxycortisol,1β-royprogesterone,dehdroepandrosteronesulfatadestriol)iththeprogreioofcy indicatingaclosecorrelationwiththedevelopmentoffetal-placentalfunction.Somehormonesincreasedinthethirdtrimester possiblydue tomaternaladaptiveregulationand maturationoffetaladrenal function.Conclusion Thedetectionresults using themethodestablishedinthisstudyrevealdynamicvariationsofsteroidhormonesduringpregnancywhichprovidesues forfurther physiological and pathological studies of these hormones.
