UXS1在肝细胞癌中的表达及临床意义

【Abstract】ObjectiveTo investigatetheabnormal expressonandclinical significanceofUDP-glucuronatedecarboxylase 1(UXS1)inhepatocelularcarcinoma(HCC).MethodsBioinformaticsanalysis was employed to preliminarilyexplore the potential functional mechanismsof UXS1in HCCandits prognosticvalue,and expresion diferencesbetween HCC tissues andadjacent tisses were verified through real-time fluorescence quantitative PCR(qPCR)experiments.ResultsAnalysis of the TCGA-LIHC database found that UXS1 was significantlyupregulated in HCC.Multivariate Coxregression analysis showed that UXS1wasan independentrisk factor,while GSVAanalysissuggested that UXS1 mightparticipateintumor progresion byregulating the PI3K-Akt-mTOR signaling pathway,andsignificantly enrichedsmall molecule metabolism, aminoacidmetabolism,andsignalreleaseinbiological processes.TheROCcurveconfirmed thediagnosticeficacyof UXS1 in distinguishing cancer from adjacent cancer ( AUC=0.96 ),and the column chart constructed based on UXS1 and clinical featuresofamultifactorial Cox model predictedthesurvivalriskat1,2,and3 years(C-index=O.6),which wasvalidated through calibration and decisioncurves.QPCR experiments showed that UXS1 was significantly upregulated in HCC( P=

0.011).ConclusionThis study is thefirst toreveal that UXS1maypromote HCC progressionthrough ametabolism-signaling pathwaysynergymechanism.Itprovidesatheoretical basis forUXS1asa prognosticbiomarkerandtherapeutictarget, and ofers new directions for exploring novel therapeutic strategies for HCC.
