枸杞多糖缓解铝致Sertoli细胞毒性的机制研究

【Abstract】ObjectiveToinvestigate thetoxicefectsof aluminum exposureonSertolicells(TM4celline)andits underlying mechanisms,and to evaluate thealleviating effectof lycium barbarum polysaccharides(LBP).MethodsTM4cells were divided into four groups:normal control (NC)group,aluminum exposure (Al)group,LBP control(NC + LBP)group, and LBP intervention (Al + LBP)group,and an aluminum toxicity model was established. Cell viability was detected by CCK8assay,apoptosisratebyflowcytometry,reactive oxygen species(ROS)levels byfluorescence microscopy,andtheprotein expressionsof glialcelline-derived neurotrophicfactor(GDNF),Catalase,superoxidedismutase2(SOD2),Bcl-2,BAX, Cleaved Caspase-9,and Cleaved Caspase-3 by Western blot.ResultsCompared with theNC group,theAl group showeda decreaseincellviability,anincrease inapoptosisrate,anincreaseinROSlevelsandthe protein expresionsofBAX,Cleaved Caspase-9,and Cleaved Caspase-3,and a decrease in the protein expresion of GDNF,Catalase,SOD2,andBcl-2(all P< 0.05).Compared with the Al group,the Al + LBP group showed an increase in cell viability,a decrease in apoptosis rate,a decreasein ROSlevelsandpro-apoptotic proteinexpresion,andanincrease inGDNFandantioxidant/anti apoptoticprotein expression (all P<0.05 ). ConclusionAluminum exposure damages Sertoli cells by inducing oxidative stress,downregulating GDNF expression,inibitingcellviability,andpromotingapoptosis.Conversely,LBPcanalleviatealumnuminducedSrtoli celltoxicitybyexerting antioxidantandanti-apoptotic efects,while maintaining GDNFexpressionandcellularviability.
