Molecular Characteristics and Expresson Patterns of the HSP20 Gene Family in Sanghuangporus baumi at Different Developmental Stages

IAN Yanting,QI Xin,LIU Hao,LIAO Fangzhou,TONG Yana (TheResearch InstituteofForestryandPomology,TianjinAcademyofAgricultural Sciences,Tianjin3Oo384,China) Abstract:Toelucidatethebiologicalfunctionsandmolecularmechanismsof theheatshockprotein2O(HSP20)genefamilyinthe medicinalfungusSnghuangporusbaumi,thisstudysystematicallyidentifiedtheSbHSP2Ofamilymembersbasedonholeenome dataandinvestigatedtheirrolesinfungalgrowthregulation.Theresultsdemonstratedthat11SbHSP2Ogeneswereidentifiedin S baumi,ncodingproteisithmolecularweightsrangingfrom17.5to8.96Daandisoelectricpoits(p)distrbutedbtwen5.07 and10.38.Phylogeneticanalysisrevealed highhomologybetweenSbHSP2Oproteinsand HSP2OproteinsfromPleurotusostreatus (average sequence identity: 76.8 % ).The promoter regions harbored 12 classes of stress-responsive cis acting elements,including ABRE,MBS,ndSRE,withtheSH2-7promoterontaningsevendistictstresselatedelements.Expressonprfligaaly sisrevealed thatthe transcription levelsof SbHSP2O-2and SbHSP20-IO wererelativelyhighinmyceliacultured for5days,with valuesof 23.20and9.023,respectively.Meanwhile,theexpresionlevelsofSHSP2-6,SHSP2-7,andSHSP20-10imyelia culturedfor9dayswere52,6.09,nd98,espectively.Colinarityalysisucoveredcromosomalhomologousgiosb tweenS.baumiiandAgaricusbisporus.Inonclusion,thisstudyprovidesthefirstsystematiccharacterzationofthemoleularfeaturesand expression regulation of the SbHSP2O gene family in S .baumii.The findings highlightSbHsP2O-1Oasakeycandidate geneforenhancingfungalstressresistance,oferingboththeoreticalfoundationsandgeneticresourcesformolecularbreedigof stress-tolerant medicinal fungi.
