改良QuEChERS/UPLC-MS/MS法检测参类中药材中杂环酰胺类杀菌剂及其代谢物残留

AbstractAn analytical method was developed for the determination of fluxapyroxad,fluopyram,fluopimomide,furametpyr,penthiopyrad,thifluzamide,furpylamin fluopicolide and the metabolin 2,6-dichlorobenzamide residues simultaneously in Ginseng(Ginseng Radix Et Rhizoma),Salviae Miltiorhizae Radix Et Rhizoma American ginseng (Panacis Quinquefoli Radix),Codonopsis Radix through improving pre-treatment process and ultra-high performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) methods.Samples were extracted with acetonitrile.Salviae Miltilorrhizae Radix Et Rhizoma extracting solution per milliliter was cleaned up with 180mg (202 MgSO4 , 30mg PSA(ethylenediamine-N-propylsilane) and 3 mg GCB (SpeedTestTM activited carbon),while the extracting solution of Ginseng,American ginseng and Codonopsis Radix were cleaned up with 20mg PSA.The purified extracts were detected by UPLC-MS/MS with Waters ACQUITY UPLC? HSS T3 column in positive ionization mode with multiple reaction monitoring and scanning, and the methanol solution containing 0.1% formic acid and aqueous solution containing 0.1% formic acid were used as mobile phase B and A respectively for gradient elute.The results showed that the 8 target compounds showed good linearity in the range of 0.0001 to 0.1mg/L with R2 over O.999. The limits of detection and limits quantitation were in the range of 0.000 01~0.000 6mg/kg and 0.0002~0.0010mg/kg ,respectively. The recovery rate was 76% to 120% at three supplemental levels of O.01,0.1O and 1.00mg/kg with the relative standard coefficient as 0.5% to 15.7% . This method had the characteristics of fast and simple pre-treatment process,good sensitivity and high accuracy,so it could be used to detect the residue of the seven heterocyclic amide fungicides and one metabolin in Ginseng and three similar herbal medicines.
