Study on Heterologous Expression of Glucose Isomerase from Pantoea dispersa of Kiwifruit Root Origin

Abstract:Toaddressthelimitations of traditionalhigh-fructose syrup production,such as lowcatalytic efficiencyof conventional glucose isomerases,this study focused on the heterologous expressionand optimization of a novel glucose isomerase, P d -xylA, derived from Pantoea dispersa isolated from kiwifruit roots. The P d. -xylA gene was cloned and expressed in Escherichia coli BL21 (DE3),folowed by systematic optimization of induction conditions including temperature and IPTG (isopropyl- β D -thiogalactopyranoside) concentration. The results demonstrated that the optimal induction conditions were and 0.1 mmol/L IPTG, under which P d? -xylA achieved the highest soluble expressonlevel.This study provided theoretical support for the development of enzyme resources with enhanced catalytic activity and industrial applicability, thereby advancing the biotechnological productionof

high-fructose syrup.
