基于高通量测序探讨芪藤消浊颗粒对慢性肾炎miRNA表达谱的影响

Abstract: Using microRNA(miRNA) high-throughputsequencing technology,this studyinvestigatedthe efects Qiteng Xiaozhuogranules(QTXZG)on miRNA expression inthe glomerular tissue rats with chronic glomerulonephris(CGN). Sprague Dawley(SD)rats were randomly divided into three groups:thecontrol group,the model group,and the group receiving QTXZG( n=3 ).A CGN model was preparedvia intravenous injection adriamycin into the tail,folowed by 30 days QTXZG administration via oral gavage.The histopathological effcts onrat glomerulartissue were assessed using hematoxylin andeosin staining(HE),and miRNA sequencing wasused to identify diferentialyexpressed miRNAs. Subsequent Gene ontology(GO)and Kyoto Encyclopedia Genes and Genomes(KEGG)enrichment analyses were used to predict related mRNAs.HE staining showed that the model group had athicker glomerular basement membrane andincreased inflammatorycellinfitration comparedwiththoseinthecontrol group,both which were significantly improved after undergoing QTXZG treatment.miRNA sequencing revealed 28 diferentiall expressed miRNAs in the model group compared with the control group,whereas the QTXZG group had 28 diferentiall expressed miRNAs compared with the model group.Atotal12 diferentiallyexpressed miRNAs were identifiedacross the three groups,andafter predicting their mRNAs,eight core mRNAs were identified.GO analysis highlighted the negativeregulation dendritic spine maintenanceand enrichment in glutamate-gatedcalcium ionchannel activity,whereas KEGG analysis indicated the involvement theRap1,cAMP,and RAS signaling pathways.qRT-PCRresults confirmed that miRNA expresion levels in rat glomerular tisue across the three groups were consistent with the trends observed in the sequencing results.QTZG regulatesthe expression miRNAs,which may,in turn,affect the Rap1,cAMP,and RAS signaling pathways,thus playing a potential role in CGN prevention and treatment.
