Abstract:Combined fingerprinting and comprehensive assessment of activity techniques were used in this study to determine thequaity control of Jingfang granules.Separation wasperformed usingan Agilent TC-C18 chromatographic column ( 4.6mm×250mm , 5μm ),gradient elution performed using an acetonitrile- .0.8% formic acid aqueous solution,and ESI-TOF/mass spectrometer was used for analysis.Thechemical constituents of Jingfang granules wereidentified through the precise molecular weightinformationof the parent anddaughter ionsof compoundsandby consulting references. Moleculardocking wasusedtoexamine thebondingabilityof the following fivechemical componentstoneuraminidase (NA): prim-O-glucosylcimifugin(POG),ferulicacid,hesperidin,naringin,and sec-O-glucosylhamaudol(SOGH).The antioxidant activityof thefivecomponentswasevaluatedbyanABTSfreeradical scavenginginvitroexperiment. Fingerprintingandquantitative assaywere used toevaluatethequaityof Jingfang granules.Fromtheresults,twenty-four chemicalcomponents wereidentified within the Jingfanggranules.Molecular dockingresultsrevealed the high binding capacity of NA to POG,ferulic acid,naringin,and SOGH,with a minimum recorded binding energy of .Free radicalscavenging invitroexperiments revealedthatal thefivecomponents exhibitedcertainantioxidantactities.The compositional similarityof diferent batches of Jingfang granuleswas foundtoberelativelyclose,but there were some diferenes inthecontentof thefivecomponents,whichmaybeatributedtodiferences inthesources of Chineseherbal medicines.This study provides methodological andconceptual supportforoverallqualityevaluationstudies of Jingfang granules.
