Abstract: Objective To analyze the expression LRP5 in glioblastoma(GBM) and explore the effect knocking down LRP5 on cell proliferation,migration abilities and MAPK signaling pathway. MethodsThe GEPIA database was used to analyze the diference in the expression LRP5 between GBM and adjacent tissues. The cancer and adjacent tissues and sections patients diagnosed bypathologywere collected,and the bioinformaticsresults were verifiedby immunohistochemistry and Western Blot. Small interfering RNA(siRNA) technology was used to knock down LRP5 for cell function experiments to observe the effects LRP5 on the proliferation, migration ability and MAPK signaling pathway GBM cell lines. ResultsBioinformatics analysis found that the expression LRP5 was increased in GBM tissues and was positively correlated with Ki-67.The results clinical tissue sample verification were consistent with the results bioinformatics analysis. The results cell experiments showed that knocking down LRP5 significantly inhibited the proliferation and migration ability GBM A172 cell line,increased the proportion cells in the G2/M phase,and inhibited the JNK and p38 signaling pathways.
ConclusionsLRP5 may promote the proliferation and migration GBM by affecting the cell cycle and activation the MAPK pathway, indicating its value as a potential molecular target for GBM.
