A coupled diffusion-based model of interaction between tumor metastasis and myeloidderived suppressive cells

Abstract: Objective Toexplore thekeyroleof myeloid-derived suppresive cels (MDSCs)inpre-metastaticniche (PMN)and analyzetheir interrelationships withthemain components inthe microenvironment usingamathematical model.Methods Mathematical descriptions wereused tosystematicallanalyzethefunctions of MDSCs intumor metastasisand elucidate their associationwiththemajorcomponents (vascularendothelialcells,mesenchymal stromalcels,andcancer-associated macrophages)contributing tothe formationof the pre-metastaticmicroenvironment.Based ontheformation principleof the pre-metastaticmicroenvironmentoftumors,thekeybiologicalproceseswereassumedtoconstructacoupledpartial differential difusionequation model.The existenceanduniquenessof the model solutionswere investigatedusing approximationmethods,thequalitativetheoryofpartialdiferentialequationsandBanach'simmovablepointtheoremand numericalsimulationswerecariedoutbydiferentialnumericalmethodstoverifythereliabilityandacuracyofthemodel. ResultsThe existence anduniquenessofthelocalandoverallsolutionsof themodelwere provedusing theapproximation method,thequalitativetoryofpartialdierentialequatinsandanach'simmoablepointteorminombinationwiththe regularityestimationof thelocalsolutionsandtheembeddinginequality.Numericalsimulationresultsfurther validatedthe reliabilityof the modeland demonstratedthe importantroleof MDSCs inthe pre-metastatic microenvironmentof tumors, especiallyinangiogenesisand immunosuppresion.Conclusion This studyreveals theimportantfunctions ofMDSCsinthe pre-metastatic microenvironmentof tumors through mathematical modelingandnumerical simulation,which providesan importanttheoreticalbasisfor understandingthemechanismoftumor metastasisanddevisingcancertreatmentstrategies.
