Abstract: ObjectiveTo investigate the impact of prenatal fear stress on placental aminoacid transportandemotion and cognitiondevelopment inofspringrats.MethodsThirtypregnantWistarratswererandomizedequallyintocontroland fear stress(inducedusinganobservationalfootshockmodel)groups.Ineachgroup,placentalandserumsampleswerecollected from6damsongestationalday2O,andtheremainingratsdeliverednaturallyandtheofspringratswereraisedunderthe sameconditionsuntil8weeksofage.Emotionalandcognitiveoutcomesoftheoffspringrats wereasessed withbehavioral tests,andplacental structurewasexaminedusingHEstaining.Bioinformaticsanalysiswasused toidentifydiferentially expresed placental transporter genes under fear stressThe expresionsof system AandsystemLaminoacid transporters, alongwithotherspecializedtransporters,weredetectedusingqRT-PCRandWesternbloting.Fetalserumaminoacid concentrations weredeterminedbyHPLC.Thecorelationsbetweenfetalaminoacid levelsandbehavioraloutcomesof the ofspringratswereanalyzed.ResultsThedams withfearstressshowedreducedopen-fieldactivityandincreasedfreezing behaviorwithsignificantlyecreasedplacentalweight,fetalweight,andfetal-toplacentalratio.ioinformaticsaalyis revealed28diferentiallyexpressedtransportergenes involvedmainlyinaminoacidtransport.Inthefearstressgroupfetal serumaminocidlvelsresignificantlylweeddS38al,c4a1,43a,a8,6a6,lcaandSaad proteinexpressions werealldownregulated.Theofspringratsinfearstressgroupexhibiteddecreasednovelobjectpreference andspontaneousalternationwithreducedopenarmexplorationandincreasedimmobilityinemotional tests.Lowerearly-life aminoacidlevelswasfoundtocorrelatewithimpairedadultcognition.ConclusionPrenatalfearstressinratsimpairs placentalaminoacidtransporterexpressonandreducesfetalserumaminoacidlevels,potentiallycontributingtoo-term cognitive deficits in the offspring rats.
