Abstract: Objective To exple the mechanism of Qixiong Zuogui Granules (QXZG) for enhancing synaptic plasticity in aging rats.MethodFortySDrats wererandomized intocontrol group,agingmodelgroup,donepezil treatment group,and QXZG treatmentgroup(n10).Exceptforthcontroats,allteatsweresubjectedtodailyintraperitonealijectionofDgalatoe for8consecutive weeks toinduce brainaging,and donepezil hydrochlorideandQXZGsuspension were administeredby gavagedurigmodeling.Afterteterenions,teatswereevauatedforgenralonitions,eavioralngesidtive stressindicators,ppocaalpathologiesandexpressionsofthebaideivedurotroicactor(D)/tyoieise receptorB(rkB)pathwayp16andsyapticplasticity-associatedprotens.ResultsTeratsinthemodelgroupbied obviousagingphenotypessuchasyellowingof theteethandhair,bodyweightlossandimpairedlearingandmemory abilities,withdecreasedserumSODandGSH-PxactivitiesandincreasedserumMDAlevel.Theratmodelsalsoshowed obviouspathogcalgcdissiesndted16oteepsiineppoaloith significantlydecreased expressionlevelsof BDNF,TrkB,CREBandsynaptic plasticityproteins SYN,GAP43,andD95. Treatment with QXZG allviatedtheagingphenotypes intheratmodels,improved their learningand memoryabilitiesand pathologicalcanges intheppocapalCAegion,ducedoxdativestressandp16proteiexpresion,andprootedthe expresions of the BDNF/TrkB pathwayproteinsand synaptic plasticity proteins.Conclusion QXZG enhances synaptic plasticityand reduces oxidative stressin aging rats possibly by upregulating the BDNF/TrkB signaling pathway proteins, thereby delaying brain aging and improving learning and memory abilities of the rats.
