Ziwuliuzhu acupuncture modulates Glu/GABA-Gln metabolic loop abnormalities in insomniac rats

Abstract:ObjectiveToinvestigatethetherapeuticefectofZiuuliuzhuacupunctureinaratmodelofinsomniaandits regulatoryeffecton theglutamicacid(Glu), γ -aminobutyricacid(GABA)-glutamine (Gln) metabolic loop.MethodsFortymale SDratswererandomlyassignedtoontrolgroupmodelgroup,NajiagoupndNaigroup(n10).Ithelatter3roat modelsof insomnia wereestablishedbyintraperitoneal injectionsof p-chlorophenylalanineandverifiedusingasodium pentobarbital-induced sleeptest.After modeling,theratsinNajiaandNazi groupsreceivedacupuncturefor7daysat specificallychosensetsofacupointsbasedontheZiwuliuzhurationaleintraditionalChinesemedicine.Pathologicalchangein the hypothalamic tisse of therats were examined withHEstaining,andthe levels of Gluand GABA in the hypothalamus were determined withhigh-performance liquid chromatography (HPLC)-mass spectrometry (MS)/MS.Immunohistochemistry was used to detectthe expressions of GABAA receptors (GABAARs)in the hypothalamus,and the expression levelsof glutamatedecarboxylase (GAD65/67)andglutamine synthetase (GS)were determinedwith Western bloting.Results Compared withthemodel group,therats inNajiaandNazi groups exhibited decreased Glulevelsand GABAAreceptor expresionand increased GABA levels with a decreased Glu/GABA ratio in thehypothalamus. Ziwuliuzhu acupuncture significantlyincreasedtheproteinexpresionsofGAD65andGAD67andloweredtheexpressionofGSinthehypothalamus intheratmodelsof insomnia.Conclusion Ziouliuzhuacupunctureproducessedativeandhypnoticefects inratmodelsof insomniaposiblybyregulatingGluand GABA-Glnmetabolism torestoretheexcitatory/inhibitorybalancebetween Gluand GABA.
