Thegene expression profile of Vibrioparahaemolyticus exposed to acute ammonia*

[Abstract]Objective:To investigatetheefectsofacuteammoniaonthegeneexpressonandfunctionalphenotypesof Vibrio parahaemolyticus(VP).Methods:Usingacontrol groupwithcultureconditionsthatdidnotincludeNH4Clandanexperimental group with NH4Cl added at a final concentration of 71.68mg/L (simulating acute ammonia exposure),the RNA-seq assay was employedtoanalyzethegeneexpressionprofileofVP.Additionall,aseriesofphenotypicexperiments-includingcrystal violetstaining,motilityrelatedphenotpeassessment,andcyclicdiguanylate(c-di-GMP)contentmeasurement-werecoucted toexploretheimpactofacuteammoniaonthesefunctionalphenotypes.Results:Acuteammoniaexposureled to significant alterations inthetranscription levelsof 661 genes,including 379downregulated genesand282upregulatedgenes.These geneswereprimarilyconcentrated incellularmetabolicpathways,particularlythosecloselyrelated toaminoacidtransport andmetabolism,as wellasenergyproductionandconversion.Furthermore,acuteammoniaalsoregulatedthetranscriptionof several genes associated with biofilm formation, c -di-GMP metabolism,flagellar function,and key virulence factors.However, acuteammoniadidnotexhibitregulatoryefectsonthefunctional phenotypesthatcorrespond tothesegenes.Conclusion: Acuteammonia induced extensivetranscriptional changes inalargenumberof genes within VP,especiallthoserelated to metabolism.However,ithadnoregulatoryefectonfunctional phenotypessuchasbiofilmformation,c-di-GMPmetabolism, or motility.
