Abstract:Classical swine fever(CSF) caused byclassical swine fever virus (CSFV),isa highly contagious and lethal viral disease inswine,forwhich vaccinationremainsacritical preventivestrategy,antibody levelsisan important indicator for evaluating theimmuneefectof immunizedpig herds.In thisstudy,therecombinantE2Y proteinof CSFV wassuccessfully expressed using a prokaryotic expression system.The purified E2Y protein served as a coating antigen,andan indirectELISA method for detecting CSFV-specific antibodies was established throughthe systematicoptimizationof experimental conditions.Results indicated thattheoptimalcoating concentrationof the E2Y protein was 0.25μg/m L.The assay demonstrated no cross-reactivity with positive sera against common swine viruses andexhibitedhigh specificity,sensitivity,andreproducibility.Theconcordance between thismethod anda commercial ELISA kit reached 91.67% .This studysuccessfullydeveloped anindirect ELISA for CSFV antibody detection,providingatechnicalbasis forfurtheroptimizationanddevelopmentof CSFVantibodydetectionreagents.
