Response characteristics of DNRA bacteria in paddy soil under wet and dry alternation

Abstract:ThispaperaimsatstudyingtheresponseofDNRAbacteriacommunitystructuretodryandwetalternationinpaddysoils with diferentfertirsndtoalyteesposecaracteristisofsoilAbcteriatostureanagementUsingpddysoilrolg term positioning test site,three treatments of single application of chemical fertilizer(NPK), 30% organic fertilizer + 70% chemical fertilizer(OM),andnofertlizer(CK)ascontrolwereseletedwithincubationexperimentsunderalteratingetanddry(cotuous weting,continuousfloding,andalternatingwetanddry)SoilDNAwasextractedafter23dofincubation,andhigthroughput sequencingoftheDNRAfunctionalgenenrfAwasperforedtonvestigatetheresponseofteDNRAbacterialcommunitystructuretowet anddryatemation.Theresultsshowedthat:thecompositiosandrelativeabundanceofRAbacterialcommunitiesinpaddysoilsere changedtosoeetentAttemlevel,etdryalteatiodecreasedtelatiebundaneofseudomoadotaVeucobota andChloroflexotaandincreasedtherelativeabundanceofAcidobacteriota.Continuousfloodingfavoredtherelativeabundanceof ChloroflexotabutdecreasedtherelativeabundanceofActinomycetota.TherelativeabundanceofActiomycetotawashigherunderthe continuous wetting treatment. Community α -diversity analysis showed that wet-dry alternation decreased Chaol index,Shannon index and SimpsonindexofsoilDNRAbacteria,utShaonindexandSimpsonindexwerehgherincontiuouslyfloodedsolthaninwetratmnt. Underthesamewatermanagementconditions,therewasnosignificantdiferencein ShannonindexofDNRAbacteriainpaddysoil betweendiferentfertilizationtreatments.TheresultsofprincipalcomponentanalysisshowedthatthecomunitystructureofDNRA bacteriainpaddysoil differed significantly among diffrent moisture treatments.The communitystructure and α- diversity of DNRA bacteria in paddy soil responded to the moisture changes,but the response of α- diversity to different fertilizers was not significant.We hypothesize thatthe influence of moisture managementon DNRA bacteria was greater thanthatoffertilizer management.
