帕金森病患者血清miR-4-3p、miR-4-3p表达水平与病情严重程度的关系及其早期诊断价值

Abstract: Objective To analyze the relationship between the expression levels serum microRNA-4-3p and miR-4-3p with the severity Parkinson′s disease (PD), and their early diagnostic value. Methods From January 08 to December 03, 96 PD patients admitted to were selected as the PD group, and 06 healthy individuals were selected as the control group. According to the severity 96 patients, they were grouped into the early stage group (I-II stage, n=59 )and the mid to late stage group (III-V stage, n=37 ). RT-qPCR was applied to detect serum levels miR-4-3p and miR-4-3p. Spearman method was used for correlation analysis. The influencing factors PD severity were analyzed using multiple Logistic regression analysis. The efficacy serum miR-4-3p and miR-4-3p in diagnosing PD and its severity was analyzed by receiver operating characteristics (ROC). Results Compared with the control group, the serum levels miR-4-3p and miR-4-3p were lower in the PD group (P<0.05) . Compared with the early stage group, there were a longer disease course, higher level homocysteine (Hcy) and lower levels serum miR-4-3p and miR-4-3p in the mid to late stage group ⋅P<0.05 ). Spearman analysis showed that serum miR-4-3p and miR-4-3p were negatively correlated with Hoehn-Yahr staging in PD patients ⋅P<0.05 ). Multivariate Logistic regression showed that disease duration ( OR=.49 ), Hcy ( OR=.56 , serum miR-4-3p OR=0.95 ) and miR4-3p OR=0.97 ) were factors influencing the severity PD ( P<0.05 ). ROC analysis showed that the sensitivity and specificity serum miR-4-3p and miR-4-3p combined in the diagnosis PD were 85.4% and 8.90% respectively. The sensitivity and specificity serum miR-4-3p and miR-4-3p in the diagnosis advanced PD were 89.6% and 79.66% respectively. Conclusion The serum levels miR-4-3p and miR-4-3p are relatively low in PD patients, and their levels are related to the severity disease. The detection serum miR-4-3p and miR-4-3p is helpful for the diagnosis and evaluation PD.
