Abstract:Lactatehaslongbeenregardedasametabolicbyproductofglycolysis.Recentstudies,however,haverevealedits pivotalrole inregulatingthe tumorimmune microenvironment (TIME).Excesslactateaccumulationdisrupts metabolicbalance, suppresses theactivityofTcellsandNKcels,andinduces thepolarizationofimmunosuppressivecels,therebyreshaping the immunelandscapeofTME.Meanwhile,lactylation,asanovelpost-translationalproteinmodification,mediates thegulation ofepigeneticeventsbymetabolicsignals,promotingtheexpreioofimmue-reatedgenes,schasD-,I-1Agl, andfurtherenhancing immune evasion.Thisreview focuses onthemetabolic-epigeneticcoordinationoflactateandlactylation inTIME,elaboratingonlactateproductionandshutling,immune-cellmetabolicinteractions,andtheclicalpotetialof targetinglactatehomeostasis.Adeeperunderstandingof thelactate-lactylationaxis mayprovidenovelmoleculartargetsand research directions for future tumor immunotherapy.
