Effects of Fully Synthetic Medium on the Production ofGibberellins throughFermentation

Abstract: Objective To reduce the cost of the fermentation medium and increase the content of the target products GA3 and GA1 , the compound medium formulation for the synthesis of gibberellins utilizing liquid submerged fermentation technology with Gibberella fujikuroi should be optimized.MethodThis study conducted an indepth investigation based on the fermentation composite medium formula of Sichuan Lomong Fusheng Technology Co.,Ltd.By adopting a rational design approach based on the principle of conservation of massconversion, the nitrogen content of the nitrogen sources,soy protein powder,and corn gluten meal in the fermentation composite medium was theoretically calculated. In conjunction with the addition of other welldefined substances,ammonium sulfate,urea,citric acid,and vitamin B1 were determined to replace the original nitrogen sources. Subsequently, the addition amounts of these substitute substances wereoptimized basedon experimentaldata,ultimately leading to the development of a fullysynthetic fermentation medium formula.ResultsThe optimaladditionamounts for the fully synthetic medium were: ammonium sulfate 4.000g/L ,urea 0.033g/L, citric acid 1.000g/L , and vitamin B10.077g/L. which effectively replaced the soy protein powder and corn gluten meal inthe compound medium as nitrogen sources.

Conclusion Under the conditions of this fully synthetic medium,the production of gibberellin GA3 ina 50L -scale fermentation reached 6.167g/L ,representing an increase of 8.9% compared to the production of 5.663g/L using the compound medium. In comparison to the production of 0.567g/L utilizing the compound media, the production of gibberellin GA1 increased by 55.7% to 0.883g/L . At 4.788 yuan per batch, the wholly synthetic media was 26.42% less expensive than the compound medium, which cost 6.5O7 yuan per batch.
