Construction Lipid Droplet Specific FluorescentProbesand Studies on Cell Imaging

Abstract: ObjectiveTo develop a novel fluorescent probe with high selectivity for lipid droplets,excellent imaging ability,and easysynthesis.MethodDesignand synthesize afluorescent molecular probe LD-TPA with a D - π - A structure, and characterize its structure by 1H NMR, 13C NMR, and HRMS. Study the sensitivity the polarity response probe LD-TPA through spectral testing experiments.Study the stability probeLD-TPA under different pH environments.Select common inorganic salts,reactive oxygen species,amino acids,and other biomolecules as interferents to studythe anti-interference properties probe LD-TPA.Probe LD-TPA's cytotoxicity was examined using the MTTassay,and its capacity to localize to lipid droplets was examined by theuse laserconfocal microscopy for zebrafish,oleic acid stimulation,and organelle co-localization assys.ResultThe probe LD-TPA exhibits the AIE effect in a mixed solvent Dioxane /H2O and produces a regular fluorescent response to solvent polarity.The probe LD-TPA exhibits excellent characteristics such as stronganti-interferenceability,pH stability, and low cytotoxicity.Simultaneously,the probe LD-TPA has amazing dynamic imaging characteristics,including good lipid droplet localization and targeting abilities as well as thecapacityto track changes in lipid droplets cells upon stimulation byvarious external conditions.In adition,the probeLD-TPA can specifically target lipid droplets in zebrafish,therebyachievingdynamic tracking ipid droplets in living cels.ConclusionThefluorescent probe LD-TPA has potential applications in the diagnosis and treatment lipid droplet related diseases,and provides effective probe compounds for dynamic tracking lipid droplets in cells.
