皖南蝮蛇抑瘤组分-I通过调控RAI14抑制顺铂耐药胃癌细胞的增殖与侵袭

Abstract:ObjectiveToevaluatetheinhibitoryefectofantitumorcomponent-IinAgkistrodonhalysvenom(AHVAC-I)on proliferationandmigrationofcisplatin-resistant gastriccancercellsandexploretheunderlyingmechanism.Methods Cisplatin-resistantMKN-28(MKN-28/DDP)cellswereobtainedbycontinuousexposureofMKN-28cellstostepwiseincreasingconcentrationsofcisplatin.MKN-28/DDPcelsweretreatedwithdiferentconcentrationsofAHVAC-Landthe changes inproliferation,migratioandvasionofthecelsereexamiedwitholonforingaCCK-8ssaoudhealing assayand Transwellassy. Westernbloting was performed toexamine theeectof AHVAC-Ionexpressionsof epithelial-mesenchymaltransition (EMT) markersof MKN-28/DDPcels;thechanges in proteinand mRNA expresionof retinoic acid induced14(RAI14)was detected with Western bloting and qRT-PCR. Results Treatment with2,4,and 8μg/mL AHVAC-Isignificantlyinibitedproliferative,migratoryandinvasionabilitiesandreducedthe expresionsofEMmarkers in MKN-28/Dcells.adih-28cells,-8/cellssodaniceasdexeionof14,ihas significantlyloweredafter treatmentwithAHVAC-1.SupplementationwithexogenousRAI4obviouslyatenuated the inhibitoryefectofAHVAC-IonproliferationandmigrationofMKN-28/DDPcels.ConclusionAHVAC-I decreases proliferation and invasion of MKN-28/DDP cells by downregulating RAI14 expression.
