颈椎病颈椎终板软骨的转录组学特征及针刀干预对FGF18/Akt 轴关键分子表达的影响

Abstract:ObjectiveToexplorethetranscriptomiccharacteristicsoftheendplatecartilageoftheervicalintervertebaldiscn cervicalspondylosis (CS)andtheefects ofacupotomyonexpressionsoffibroblast growthfactor18(FGF18)/proteinkinase B (Akt)axiskeymolecules.MethodsTranscriptomicanalyses wereperformedusingbioinformaticsmethodsbasedontheGEO database.Inalepimnt,24eaandbitsredodalltotol,oelto groups.Inthelatter twogroups, CSmodels wereestablished followed7dayslaterbyacupotomyintervention (nceaweek for 3weeks)ornoparticulartreatment.Histopathologicalchangesandcellapoptosisintheintervertebraldiscswereexamined withHEstainngandUNELassayndthemRNAexpressonsofGF18,GFR3,andAktintheintervertebraliscswere detected using RT-qPCR;the localizationand expressions ofFGF18,p-Akt,and Akt proteins were examined with immunohistochemistry.ResultsTheendplatecartilageinCSexhibitednumerousdiferentiallyexpressedgenesenrichedin thePI3K-Aktsignaling,calciumsignalingandRap1signalingpathways.CSabbitsshowedobviouschangesinthecevical spinecurvatureandjointdegenerations,changesincervicalintervertebraldisctexture,thinningoftheannulusfibrosus, shrinkageorevenabsenceofthenucleuspulposus,andincreasedapoptoticcelsintheendplatecartilage,whichwereall obviouslyalleviatedafteracupotomy.Nosignificantdiferences werefoundinthemRNAexpressionsofFGF18,FGFR3and Aktin thecervical intervertebral discs among the3 groups.Acupotomy significantlyincreased FGF18 and p-Akt protein expresionsandreducedtheAkt/p-AktratiointhecervicalendplatecartilageofCSrabbits.ConclusionThecervicalendplate cartilageofCSshownumerousdiferentiallyexpressed genes.Acupotomymaydelaydegenerativechangesof the intervertebral discsandimproves CSbyactivatingtheFGF18/Aktaxistoreduceapoptosisofendplatecartilagecells.
