Abstract Objective:To explore the effect of Sequoiol on induced myocardial cell injury by regulating the Hippo signaling pathway. Methods:Human myocardial cels AC16 were divided into the control group, H2O2 group,low,medium,and high-dose Sequoiol groups (L-SequoiolgropequoigroupHSequgroup)ndU-goupEdUsainigsaplidtetectC1celifatio Theapoptosis of AC16 celswasdetectedbythe Hoechst 33258 staining method. γ -H2AX immunofluorescence assaywasapplied to detecthelevelofDNAdamageinAC16cels.Theenzye-linkedimmunosorbentassay(ELISA)methodwasusedtodetectthelevelsof superoxide dismutase(SOD)andreactiveoxygen species(ROS)in theserumof AC16cels.Theproteinimmunobloting method(Westem Blot) was used to detect the protein expressions of YAP1,p-YAP1,TAZ, γ+12AX ,B-cell lymphoma 2(Bcl-2),Bcl-related X protein(Bax), andCaspase-3 in AC16 cels.Results:Compared with the control group,the EdU positive cellrate,SOD,and p -YAP1,Bcl-2protein expression of AC16 cells in the H2O2 groupdecreased,the apoptosisrate,DNA damage,ROS,YAP1,TAZand ,Bax,and Caspase-3proteinexpression levels increased( P<0.05 );comparedwiththe H2O2 group,the EdU positive cell rate,p-YAP1,SOD,and Bcl-2 proteinexpressonincreasedintheLequoilgroup,SequilgroupandHSeqoiolgroup,teapoptosisrate,damage and YAP1,TAZ, γ -H2AX,Baxaspase-3proteixpreinvelsreaedadastecetratioofoolieadih gradually increasing ordecreasing trend( P<0.05 ;the EdU positive cell rate,SOD and p-YAP1,Bcl-2 in the XMU-MP-1 group were lower than those in the H-Sequoiol group,the apoptosis rate,DNA damage,ROS and YAP1,TAZ, γ+H2Ax ,Bax,Caspase-3protein expression inthe H-Sequoiol group were higher than those in the H-Sequoiol group( P<0.05 ).Conclusion: Sequoiol can inhibit H2O2 induced myocardial cellinjury,and its mechanism relates regulating the Hippo signaling pathway.
