AbstractObjective:ToxploretheefectofMingjunPowderonatheroscleroticratsbasedonteTollikereceptor4(R4)eloid differentiation protein 88(MyD88)/nuclear factor- κB(NF-κB) signaling pathway.Methods:The atherosclerotic model of rats was established byhigh-fat feed combinedwith vitamin method.The rats were divided into the blank group,the model group,the Western medicine group[atorvastatin 1mg/(kg⋅d)] ,the low-dose Mingjun Powder group[2.35 mg/(kg⋅d)] ,the medium-dose Mingjun Powder group[4. 70mg/(kg⋅d)] and the high-dose Mingjun Powder group [9.40mg/(kg⋅d)] ,with 10 rats in each group.After gavaging for eight weeks,the levels of serum totalcolestes)otit detected.Hematoxylineosin(HE)stainingwasusedtoobservethehistologicalmorphologicalchangsofthethoracicaortainratsTe expressions of TLR4,MyD88 and NF- κB mRNA in the aorta of rats were detected by reverse transcription polymerase chain reaction (RT-PCR).The expressionsof TLR4,MyD88,and NF-κB proteinsintheaorta of ratsweredetected byWestern Blot.Results:Compared withthe modelgroup,thelevelsofserumTC,TGandLDL-Cin theWestemmedicinegroup,themedium-dseMingjunPowdergroup, and the high-dose Mingjun Powder group decreased,and the level of HDL-C increased P<0.05 or P<0.01 ).The expressions of TLR4, MyD88, NF- κB mRNA and protein in the aortic tisues ofrats in the Westem medicine groupand each dose Mingjun Powder groupsall decreased( P<0.05 or P<0.01 ).Conclusion:Mingjun Powder might playananti-atherosclerotic efect by inhibiting the TLR4/MyD88/NF-kB signaling pathway.
