AbstractObjective:ToobservetheefectofPanaxnotoginsengsaponins(PNS)onmyocardialischemia-repefusion(I)injuryand exploreitsmechanismrelatedtotheregulationofcardiomyocytepyroptosis.Methods:EighteenadultmaleSDratsweredividedintosham group,IR groupandPSgroupbyrandomnumbertablemethod,with6ratsineach group.Themodelwaspreparedbyligatigtheleft anteriordescendingcoronaryarteryAD)ofratswithischemiafor30minutesandreperfusionfor2hours.Theatsinthesamgroupand the IR groupwere intraperitoneally injectedwith normal saline( 10mL/kg) ,while those in the PNS groupwere intraperitoneally injected with (20 PNS(50mg/kg) 30 minutes before modeling.The degree of myocardial injury was observed by hematoxylin-eosin(HE) staining.The expresionofcsteineaspartatespecicprotease-1Caspase1)asdetectedyimmunoistochemistryeexpresiosofCaspse1 andgasdermin D(GSDMD)proteincontentsinmyocardialtisseweredetectedbyWesternBlotResuts:Comparedwiththeshamgroup, thedegree ofmyocardialinjuryintheIRgroupwasaggravated,andtheproteinexpressionsofaspase-1andGDinceased P< 0.05).ComparedwiththeIRgroup,thedegreeofmyocardialinjuryinthePNSgroupwasaleviatedandtheproteinexpressionsof Caspase-1 and GSDMD decreased( ⋅∠1.5′ .Conclusion:Panax notoginseng saponins could alleviate myocardial iR injury in rats,and its mechanism might relate to the inhibition of cardiomyocyte pyroptosis.
