Research progress on the mechanisms of traditional Chinese medicine regulating NLRP3 inflammasome to interveneinrenalfibrosis

ABSTRACTRenalfibrosis(RF),characterizedbyglomerulosclerosisandtubulointersitialfibrosis,isacentralpathological process inchronic kidneydisease(CKD).The nucleotide-binding domain leucine-richrepeatand pyrindomain-containingreptor 3(NLRP3)inflammasomeiscloselylinkedtotheoccurrenceandprogressionofRF.Thisreviewsystematicallyelucidates the mechanismsoftheNLRP3 inflammasome inRF,andsummarizes thecurrentresearchontheinhibitionofNLRP3 inflammasome activationbysingleChineseherbs,activecomponentsof traditional Chinesemedicine,Chineseherbalcompounds,andChinese patentmedicinesforthepreventionandtreatmentofRF.Theexistingstudieshavedemonstratedthat singleChineseherbs (Campanumoealancifolia,Dioscoreazingiberensis),activecomponentsof traditional Chinesemedicine(moroniside, liquiritigenin,rosmarinicacid,magnoflorine,fucoidan,etc.),Chineseherbalcompounds(Bushen huoxue formula,Tongluo yishendecoction,Shizhi formula,Bixie fenqingdrink),and Chinesepatentmedicine(Suyin jiedu granule)caninhibitthe activationoftheNLRP3inflammasome tocounteract inflammatorydamage.Thisafectsmultiplepathways (NLRP3/caspase-1/ gasderminD,NLRP3/interleukin-1β/Smad,etc.)andrelatedupstreamanddownstreamtargetsofitsactivation,effectively reducingpyroptosis,mitigatingoxidativestress,promotingmitochondrialautophagy,nhibitingfibroblastactivation,andreducing excessive extracellular matrix deposition,thereby exerting anti-RF effects.
