【Abstract】Objective:To explore the regulationof monocyte subsets by Xuanfei Tongluo Prescription through the NLRP3 signaling pathway to improve lung injurycaused by toxins in community-acquired respiratory distress syndrome(CARDS). Methods: Recombinant CARDS toxins were prepared.Forty mice were selected. Ten of them were the normal control group and were injected with thesamedose of PBS.The remaining 3O mice were injected with 40 μL of PBS containing 50μg of CARDS toxin into the trachea to create a model. They were randomly divided into model group,Xuanfei Tongluo Prescription group and azithromycin group according to the random number table,10 mice per group.The mice were administered by gavage and their blood was colected from the abdominalaorta 24 hours later. The changes in the proportion of peripheral blood mononuclear cell subsets(Ly6C + )were detected by flow cytometry.The pathological changes of lung tisse were observed by HE staining,and the lung tisse injury score was evaluated.The expressions of interleukin(IL) -1β and 1L-18 in peripheral blood were detected by Real - time PCR in lung tissue and enzyme-linked immunosorbent assy.The expression of NLRP3 in lung tissue was detected by Western Blot.Results: Compared with thatof the control group,the HE injury score of the model group was significantly increased,the proportion of Ly6C+monocytes,the expressons of inflammatory factors IL-Iβ and 1L-18 in lung tissue and peripheral blood were significantly increased,andthe expresionof NLRP3 in lung tissue was significantly increased.Compared with the model group,the Xuanfei Tongluo Prescription group and the azithromycin group could reduce the pathological injury score,the proportion of Ly6C + monocytes,the expressions of inflammatory factors IL-1β (20 and 1L-18 in lung tissue and peripheral blood,and the expression of NLRP3 in lung tissue. Compared with the azithromycin group,the Xuanfei Tongluo Prescription group could reduce the expresson of inflammatory factors IL-1β and 1L-18 in lung tissue. Conclusion: Xuanfei Tongluo Prescription can improve lung injury caused by CARDS toxins by reducing the proportionof monocyte subsets,inhibiting the NLRP3 pathwayand regulating inflammatory responses.
