AbstractThis study established18 natural distribution sites and plots in the Greaterand Lesser Khingan Mountains from 2021 to 2022,collcting 50 samples of Salix arbutifolia.The modified CTAB method was employed for genomic DNA extraction from leaves.Through ISSR-PCR amplification,10 primers were identified,and their optimal annealing temperatures were determined.The results indicate that the extracted genomic DNA meets quality standards.The genetic diversityof Salixarbutifolia populations is relatively high,with an average polymorphic band percentage of 91.91% at the species level and 92.22% overall, indicating substantial genetic variation.The Shannon information index and Nei'sgene diversity index peaked at an elevation of 548m .The genetic differentiation coefficient (GST) was calculated at 0.2303, suggesting significant gene flow among populations.The five natural populations exhibited close genetic relationships,with genetic similarity ranging from O.8867 to 0.9651 and genetic distance from 0.0355 to 0.1203.The 29 individuals were categorized into four major groups.The findings conclude thatthe genetic diversity of Salixarbutifoliaarises from genetic variation influenced by itsbreeding system and seed dispersal mechanisms,while the impact of geographic isolation appears minimal.
